You can do it either way - or actually you can also wait until it's fully cured, then wash and sand the surface before next coat.

And which is best? Well I think it depends on the situation. Sometimes it's good to wait until the first layer is tacky - theyn you won't accidentally move the bottom layer when spreading epoxy and glass. In some cases it's easy to go and put several layers in same time. So I would say it depends on a situation and personal preferences. Try both and see what works for you :)
